GRAM STAIN
Intended Use:
Grams stain kit is use for differentiation of bacteria on basis of their gram nature.
Principle:
Gram staining is a common technique used to differentiate two large groups of bacteria based on their different cell wall constituents. The Gram stain procedure distinguishes between Gram-positive and Gram-negative groups by coloring these cells red or violet.
Kit components:
Reagent bottles
Kit insert.
Physical Appearance:
Gram crystal violet clear dark violet colored liquid without any particles.
Gram Decolorizer clear colorless liquid without any particles.
Gram Iodine clear dark yellow liquid without any particles.
Gram Safranin clear dark amber liquid without any particles.
PROCEDURE
1. Prepare a thin smear on clear, dry glass slide.
2. Allow it to air dry and fix by gentle heat.
3. Flood with Gram's Crystal Violet for 1 minute.
4. Drain the stain.
5. Flood the smear with Gram’s Iodine. Allow it to remain for 1 minute.
6. Decolorize with Gram's Decolorizer until the blue dye no longer flows from the smear.
7. Wash with distilled water.
8. Counter stain with Safranin. Allow it to remain for 1 minute.
9. Wash with water.
10. Allow the slide to air dry.
11. Examine under oil immersion
