Product introduction
2 × SYBR Green PCR is a special reagent for real time PCR by SYBR Green chimeric fluorescence method. The chemical modification method containing the activity of Taq completely closed at room temperature can start HS Taq DNA polymerase, which can effectively inhibit the nonspecific amplification caused by primer non-specific annealing or primer dimer under low temperature conditions, and improve the specificity of amplification reaction. After special preparation, the optimized qPCR buffer greatly improved the amplification efficiency and detection sensitivity of qPCR reaction. It can get a good standard curve in a wide quantitative region and accurately quantify. The reagent is compatible with most manufacturers' fluorescent quantitative PCR instruments, such as Applied Biosystems, Eppendorf, bio rad and Roche.
Product characteristics
High specificity: the product was initiated by thermal DNA PCR, and it had amplification activity after 90 ℃ for more than 2 minutes, which could greatly improve the specificity of PCR products. Sensitivity: the optimized buffer contained in this product can detect template with low copy number and good repeatability. High applicability: This product can be used in any fluorescent quantitative PCR instrument.
Principle of reagents
The product was amplified by PCR with HS Taq DNA polymerase, which was chemically modified by the thermal promoter, and the fluorescence signal intensity was detected by the product chimed with SYBR green.
1,PCR
PCR is a method of amplification of target fragment with DNA. Through the three steps of thermal denaturation, primer annealing and DNA polymerase, a large number of DNA fragments can be amplified in a short time.
2. Fluorescence detection
SYBR Green is a double stranded DNA binding dye that binds to the small groove. After binding with double stranded DNA, the fluorescence of the DNA was enhanced greatly, the maximum absorption wavelength was 497nm and the maximum emission wavelength was about 520nm. SYBR Green I can be combined with all the double stranded DNA, and it can be detected without using probes. It has good universality and high sensitivity. However, because SYBR Green is combined with all the double stranded DNA, false positives caused by primer dimer, single chain secondary structure and wrong amplification products will affect the accuracy of quantitative analysis. In the process of quantitative instrument detection, the fluorescence changes after rising temperature can be measured, and the homogeneity of products can be analyzed by the chain breaking curve, so as to distinguish the melting peak temperature of the products and distinguish the specific and non-specific products.

matters needing attention
1. 2 × SYBR Green pcrmix was specially prepared and chemically modified with heat starter enzyme, which had higher specificity;
2. Three steps are recommended for primers with low annealing temperature or longer than 200bp;
3. Before and after amplification, special area and liquid transfer device shall be used. Gloves shall be worn for operation and frequently replaced. Do not open the reaction tube after PCR reaction. The results showed that the contamination of PCR products to the samples was minimized.
