Introduction
The HiPure FastFilter Plasmid DNA Maxi Kits combine the power of HiPure technology with the time-tested consistency of alkaline-SDS lysis of bacterial cells to deliver high-quality DNA. HiPure DNA columns facilitate the binding, washing and elution steps thus enabling multiple samples to be simultaneously processed. This system also includes a special filter cartridge which replaces the centrifugation step following alkaline lysis.
Plasmid DNA purified by this system is suitable for automated fluorescent DNA sequencing, restriction endonuclease digestion, transfection of mammalian cells, and other manipulations. Up to 1000 µg high copy number plasmid DNA or 50-500 µg low copy number plasmid DNA can be purified from 200 mL overnight culture.
Procedure
The kit is based on glass fiber filter membrane purification technology. Only need to perform simple syringe filtering-binding-washing-elution steps.
After the bacterial are collected with centrifuge, they are lysed by alkaline-SDS lysis and neutralized. The lysate passes through a syringe with a filter to remove impurities. Then the supernatant is added to the spin column for centrifugation or vacuum filtration. The plasmid DNA binds onto the glass fiber membrane. Remaining Impurities can be removed after two times wash. The purified DNA is eluted with a small amount of elution buffer or water finally.
