TTX keratinase is a specific alkaline serine protease extract from bacillus licheniformis by a liquid fermentation technique and refined by an advanced post-treatment technique.
It is a complex protease, including including reductase of disulfide bonds and polypeptide hydrolysis enzyme. Reductase of disulfide bonds acts on disulfide bond of keratin at first and break down macromolecular structure keratin into denatured protein, then denatured protein is degraded into polypeptide, oligopeptides and free amino acid by polypeptide hydrolysis enzyme, finally, keratin is disintegrated compley.
It is not only an alkaline protease, but also can break down disulfide bond, disintegrate keratin compley. So reductase of disulfide bonds in the keratinase is the key enzyme in the course of keratinolysis.

Function of protease
1.Supplement the deficiency of endo-protease for animal.
2.Increase protein digestibility in animal feed and reduce the dosage of protein in feed formula.
3.Decrease the difference of feedstuff from different area and stabilize production performance.
4.Protect the environment and lessen pollution from nitrogen.
5.Develop out some new feedstuff e.g hydrolyzing feather meal, soybean meal and blood plasma powder.
Particular function of Keratinase
1. Degrade powerfully some anti-nutritional factors to be not broken down by trypsin and chymotrypsin e.g trypsin inhibitor, agglutinin etc.
2.Increase hydrolyzing degree of protein and proportion of soluble small protein in animal feed, which is conducive to animals’ absorption, e.g rise hydrolyzing degree 20% for corn protein, 10% for meat and bone meal, 5% for feather meal and soybean meal
3.Improve the digestibility of amino acid, especially for methionine, histidine and serine, keratinase have better action.
Definition of content
One unit (U) of keratinase activity is defined as the amount of enzyme which liberates 1 μg tyrosine per minute from substrate casein at PH 10.5 and 40℃. (GB/T 23527-2009)
Applications (Adjustable according to experiment results)
1. For leather processing, it can shorten processing time, simplify working procedure and improve product quality. Recommended dosage of 100000 U/g protease is 0.02-0.1%, 20-25℃, 12-20h for retting process; 0.05-0.2%, 35-38℃, 3-6h for leather soften; 0.1-0.3%, 20-35℃, 12-20h for leather depilation.
2. For silk degumming, it may make silk lint-free and fluffy without damaging silk. After pretreatment, the recommended dosage of 100000 U/g protease is 0.8-2.4% pH9-11, 40-50℃, 30-60min.
3. For chondroitin production, it can improve yield and purity, after extracting alkali, recommended dosage of 100000 U/g protease is 0.2-0.6%, pH8-10, 40-50℃, 4-8h.
4. For heparin sodium production, it can increase uniformity of molecule and purity, after salt solution, recommended dosage of 100000 U/g protease is 2-4g per small intestine,pH9-11, 45-60℃, 4-6h.
5. For detergent production, it can remove the stains of sweat, blood, protein and milk, and get a unique detergent effect. Recommended dosage is 0.1-1.0%
6. For animal feed, it can break down protein compley and remarkable raise the availability of feed protein. Recommended dosage of 50000 U/g is 20-25g/t in complete feed
The application experiments on broiler
Divide 3000 AA broilers of 1 days into three group:
1.Control group: basal diet of corn soybean meal (CP 20.78%)
2.TestⅠ: low protein diet (CP 19.98%)
3.TestⅡ: low protein diet + keratinase 2000U/kg
The teat result as follow after 21 days:
1.Production performance of testⅠis lower than control group
2. production performance of TestⅡis higher tha control group, ADG increases 2.86%, feed conversion ratio reduces 0.07%
Above test result shows keratinase can reduce protein 0.8% and have no influence on production performance of broiler
Keratinase hydrolyzes feather meal
TTX keratinase hydrolyzes chicken feather for 6 hours at pH 10 and 55 degree centigrade, then test the content of amino acid by amino-acid andlyser and soluble protein with Bradford method.
