Intended Use
Aflatoxin M1 Immunoaffinity Column with HPLC is a quantitative method for the detection of Aflatoxin M1 in samples. Samples are purified by the column before quantitated by high performance liquid chromatography (HPLC).
Summary
Aflatoxins are toxic and carcinogenic. They are metabolites of the fungi Aspergillus flavus and Aspergillus parasiticus. There are four principle types of aflatoxin: B1, B2, G1 and G2, which are named for their respective innate fluorescent properties. Aflatoxin B1 is the most frequently encountered of the group and the most toxic one. Aflatoxins can be found mainly in cereals, nuts, cottonseed and human blood.
Aflatoxin M1 is hydroxylation metabolite of aflatoxin B1 a potent human carcinogen.
Principle
The detection is based on antigen-antibody reaction. The antibody is bound in the column.
Samples are prepared by mixing with an extraction solution, blending and filtering. The extract is then applied to the column bound with specific antibody to aflatoxin M1. At this stage, the aflatoxin M1 binds to the antibody in the column. The column is then washed to rid the immunoaffinity column of impurities. By passing methanol through the column, the aflatoxin B1 is removed from the antibody. This methanol solution can then be injected into an HPLC system.
Kit Composition
Each kit contains Aflatoxin M1 Immunoaffinity Columns of different specifications and one copy of Instruction for Use.

The Assay ProcedureFor Immunoaffinity Column
---- Take out the column, connect the bottom of the syringe and the plug of the column on the top and press to pierce through.
---- Fix the column with syringe on the Air-Pressure Controller.
---- Remove the plug on the bottom of the column and pass the treated sample solution completely through the immunoaffinity column at a rate of about 1-2 drops/second until air comes through column.
---- Pass 10 mL of deionized/distilled water through the column at the rate of 2-3 drops/second.
---- After the liquid through the column completely, change a new syringe to connect with the column, pass 2mL of acetonitrile through the column at the rate of 1 drop/second, then repeat pass another 2mL of acetonitrile through the column for the second time. Collect the eluent to a graduated tube.
---- Blow the eluent to extremely dry by nitrogen at 30℃.
---- Dissolve the dried sample using the initial mobile phase to 1mL.
---- Filter the solution through 0.22μm syringe filter to a sample bottle for HPLC analysis.
