Introduction
AO belongs to tricyclic heteroaromatic fuel, which can mark DNA and RNA. It belongs to heterochromic fluorescent dye. The dye has membrane permeability and can penetrate cell membrane to dye nuclear DNA and RNA. Therefore, AO is often used to detect DNA and RNA in cells. The binding modes of AO and nucleic acid are as follows: 1. Insertion binding. AO is embedded between the base pairs of nucleic acid double chain. This binding mode is mainly the combination of AO and DNA. Its fluorescence emission peak is 530nm, which is green fluorescence after excitation. 2. Electrostatic attraction. The positive charge AO and the single chain nucleic acid phosphate (with negative charge) produce electrostatic attraction binding, which is mainly the combination of AO and RNA. The fluorescence emission peak is 640nm which is red fluorescence after excitation. A small amount of binding will show orange or orange fluorescence. As a result, AO chimeras appear green in double stranded DNA molecules and emit orange or orange red fluorescence when combined with DNA single strand or RNA.
The AO Stain Kit is mainly composed of AO Stain Solution and AO Stain Buffer, which are often used to detect apoptosis. After staining, AO could make the nucleus show green or yellow green uniform fluorescence through the normal cell membrane, while in apoptotic cells, the chromatin is condensed or broken into fragments of different sizes, forming apoptotic bodies. AO can make them stained with dense and concentrated yellow green fluorescence or yellow green fragment particles, while yellow fluorescence of necrotic cells weakens or even disappears.AO staining is often combined with EB staining. EB staining only produces orange fluorescence in dead cells, which can distinguish normal cells, apoptotic cells and necrotic cells.
