R.B.C. DILUTING FLUID


Intended use:  


Diluting Fluid for R.B.C.


 


Principle:


RBC diluting fluid is isotonic with blood; hence hemolysis does not take place. Normal Saline also can be used. But it causes slight creation of red blood cells and allows rouleaux formation. The blood specimen is diluted 1:200 with the RBC diluting fluid and cells are counted under high power (40 x objective) by using a counting chamber. The number of cells in undiluted blood are calculated and reported as the number of Red cells per cu mm (MI) of whole blood.

Physical Appearance:


Clear colourless liquid without any particles.


 


PROCEDURE:


1.        Draw EDTA anticoagulated blood to exactly the 0.5 mark of the RBC pipette.


2.        Wipe the tip of the pipette, clean with a piece of dry gauge without touching the opening of the capillary and immerse in the freshly filtered diluting fluid.


3.        Do not insert the pipette in the bottle of counting solution.


4.        By gentle mouth suction, draw the diluting fluid steadily into the pipette to exactly the 11 mark past the bulb, rotating the pipette on its long axis to ensure thorough mixing of blood and diluent.


5.        Immediately mix the contents of the pipette thoroughly by placing the thumb over one end and shake for 1 minute.


6.        Fill the counting chamber carefully and and allow the cells to settle for 2-  3 minutes.


7.        Count cells in all 4 "W" marked corner squares.


8.        Diluted blood must be examined within 2 hours


 


RESULTS AND INTERPRETATION


Red blood cells/mm3 in the original blood = Cell counted X dilution factor / Volume counted in mm


= Cell counted X 200 / 0.02 mm3


= Cell counted X 104


 


W.B.C. DILUTING FLUID


Intended use:


Diluting Fluid for W.B.C.


 


Principle:


Glacial acetic acid lyses the red cells. Gentian violet slightly stains the nuclei of the leucocytes. The blood specimen is diluted 1:20 in a WBC pipette with the diluting fluid and the cells are counted under low power of the microscope by using a counting chamber. The number of cells in undiluted blood is reported per mm3 (µl) of whole blood.

Clear purple color liquid without any particles.

PROCEDURE:


1.        Draw EDTA anticoagulated blood to 0.5 mark in the capillary end of WBC pipette.


2.        Carefully, wipe excess blood outside the pipette by using cotton.


3.        Draw W.B.C. Diluting Fluid up to 11 mark.


4.        Mix the contents in pipette and after 5minutes by discarding few drops, fill the counting chamber and allow the cells to settle for 2-3 minutes.


5.        Focus on 1 of the "W" marked areas (each having 16 small squares) by turning objective to low powder.10X Examine under oil immersion microscope.


6.        Count cells in all 4 "W" marked corner squares.


 


RESULTS AND INTERPRETATION:


Number of WBCs / mm3 (µl) of whole blood = No. of WBCs counted X Dilution / Area counted X Depth of fluid.


 


where, Dilution=20; Area counted= 4x1 mm2 = 4 mm2; Depth = 0.1mm(constant).


No. of leucocytes / mm3 (µl) of whole blood = No. of cells counted X 20 / 4 X 0.1


= No. of cells counted X 50

CHAIRMAN AND MANAGING DIRECTOR

Mr. Mahendra Dashora holds Master of Science in Chemistry (1979) and Master of Science in Biochemistry (1981) from the prestigious Maharaja Sayajirao University of Baroda (M.S.U), Vadodara, Gujarat (India). He has over 36 years of vast experience in the field of In Vitro Diagnostic (IVD) industry. He is the promotor of Anamol Laboratories Pvt. Ltd. and is a true visionary. He has 3 years (1981-1984) of exposure at Bayer Diagnostics (now Siemens Ltd.) as Chief Scientific Officer and In-charge of Quality Assurance. 

He co-founded Accurex Biomedical Pvt. Ltd. in 1984 and took the responsibility of Director – Technical for 20 years. He was solely responsible for the development and manufacturing of all the products at Accurex. After parting with Accurex in 2004, he took over 100% shares of Anamol Laboratories Pvt. Ltd. and started the manufacturing facility at the present location.


At Anamol, his manufacturing capabilities and standardization grew in the fields of Immunology as well (Rapid Immunoassays, ELISA, CLIA, etc.)

Some of his achievements include:

1. First time indigenously manufactured enzyme based lyophilized Clinical Chemistry kits in India (1984).

2. First time indigenously manufactured liquid stable Clinical Chemistry reagents in India (1992).

3. First time indigenously manufactured Urine Test Strips in India (1984).
