Product Introduction: 

RNase A, Ribonuclease A, is an endoribonuclease that cleaves the phosphodiester bonds of single strand RNA after pyrimidine nucleotides. It attacks at the 3′ phosphate end (For example pG-pG-pC-pA-pG will be cleaved to give pG-pG-pCp and A-pG). The highest activity is exhibited with single stranded RNA. RNase A is a single chain polypeptide containing 4 disulfide bridges. In contrast to RNase B, it is not a glycoprotein. Ribonucleases do not hydrolyze DNA, because the DNA lacks 2′-OH groups essential for the formation of cyclic intermediates. RNase A can also hydrolyze RNA from protein samples. RNase A can be inhibited by alkylation of His12 and His119 and activated by potassium and sodium salts. RNAse is inhibited in the presence of heavy metal ions. RNase is also inhibited competitively by DNA.

The most common application of RNase A is to remove RNA during the preparation of plasmid DNA or genomic DNA. In addition, this product can also be used in molecular biology experiments such as RNA enzyme protection analysis and RNA sequence analysis.

 Preparation of storage solution:

Note: This is one of the commonly used methods for the preparation of RNase A storage solution. The storage solution can also be prepared by other methods according to traditional laboratory methods or reference materials (e.g., directly dissolved in 10 mM Tris-Cl, pH7.5 or Tris-NaCl solution.

 

1) 10 mg/mL RNase A storage solution was prepared with 10 mM sodium acetate (pH5.2); 

2) Heating at 100℃ for 15 min; 

3) Cool to room temperature and add 1 M Tris-HCl (pH7.5) of 1/10 volume (for example, 5 mL 10 mg/mL of RNase A storage solution containing 500 μL 1 M Tris-HCl, pH7.5);

4) Refrigerated at -20℃, storage for 2 years.

 

Note: When RNase A solution is boiled under neutral conditions, precipitation of RNase A will occur.Boil it at a lower pH, and if precipitation is observed, it may be due to the presence of protein impurities.If precipitation is found after boiling, the impurities can be removed by high speed centrifugation (13,000 RPM) and then separated for frozen storage.
